Molecular Profile: Its chemical formula is with a molar mass of approximately 3647.25 g/mol.
Extended Half-Life: The addition of DAC extends the estimated half-life to approximately 6 to 8 days in humans. Without DAC, the peptide (often called Modified GRF 1-29) has a half-life of only about 30 minutes.
GH/IGF-1 Stimulation: A single injection has been shown to increase mean plasma Growth Hormone (GH) concentrations by 2- to 10-fold for 6 days or more.
Sustained IGF-1 Levels: Mean plasma Insulin-like Growth Factor 1 (IGF-1) concentrations increase by 1.5- to 3-fold for 9–11 days after one dose. With multiple doses, IGF-1 levels can remain above baseline for up to 28 days.
Preserved Pulsatility: Unlike direct GH injections, CJC-1295 with DAC maintains the body’s natural pulsatile GH secretion, though it significantly raises the “trough” or basal levels of GH (up to 7.5-fold).
Research Findings
Body Composition: Preclinical studies in GHRH-knockout mice showed that once-daily administration could normalize body weight, length, and lean mass.
Somatotroph Proliferation: Data indicates CJC-1295 stimulates the proliferation of pituitary somatotroph cells, evidenced by increases in total pituitary RNA and GH mRNA.
Clinical Status: While initially investigated for treating lipodystrophy and growth hormone deficiency, Phase II clinical trials were terminated in 2006/2007 following the death of a trial subject from a myocardial infarction (though investigators believed the death was unrelated to the drug).
Safety and Side Effects
Common Effects: Reported side effects include injection site reactions (pain, swelling, induration), transient flushing, warmth, headache, and mild hypotension.
Regulatory Status: CJC-1295 is listed as a Prohibited Substance by the World Anti-Doping Agency (WADA) under section S2.
FDA Warnings: The FDA has identified safety concerns including increased heart rate and potential immunogenicity (immune response against the peptide).
